Screening and biotransformation optimization of a strain catalyzing 2-hydroxy-4-(methylthio) butyronitrile to methionine hydroxy analogue

Abstract

ZJB12048, a nitrilase-producing genetic engineering strain, was isolated. Under the optimized transformation conditions as followed, reaction temperature 50 ° C, initial pH value 7.0 (100 mmol/L citric acid/sodium citrate buffer solution), concentration of wet cells 0.02 g/mL and the concentration of substrates 150 mmol/L, 2 -hydroxy -4 -(methylthio) butyronitrile were transferred into methionine hydroxy analogue by the rested cells. And the obtained product was identified
as 2-hydroxy-4-(methylthio) butyric acid using HPLC, LC-MS and 1H NMR.
