Optimization of L-lysine fermentation with enzymatic hydrolysate of potato pomace as carbon source

Abstract

In this study, fermentation of L -lysine by Brevibacterium flavum with condensed enzymatic hydrolysate of potato pomace as carbon source was investigated. The composition of total sugar (the amount of condensed enzymatic hydrolysate of potato pomace), ammonium sulfate and corn steep liquor of the medium was optimized with response surface methodology. The optimal dosage of biotin and dimethyl sulfoxide (DMSO) was further analyzed based on the optimized
medium. The final optimum fermentation condition was as follows: total sugar 125.81 g/L, (NH4)2SO4 27.20 g/L, corn steep liquor 18.17 mL/L, biotin 180 μg/L, DMSO 18 mL/L. The DMSO was added after 18 h fermentation. The production of lysine reached 68.19 g/L and was 41.21% higher than
that of non -optimal culture. The potato pomace was suitable carbon source for L -lysine fermentation.
