Ketoprofen is one of the most prominent non-steroidal anti-inflammatory drugs,which was widely used in rheumatism,rheumatoid arthritis,myelitis,gout and other diseases.Most of
the commercially available ketoprofen drugs are racemic.However,the (S)-
enantiomer has therapeutic activity against inflammation and pain,while the existence of(R)- enantiomer resulted in increased physiological side effects and toxicity.Chiral ketoprofen is currently produced by
chemical synthesis.Compared to chemical methods,biosynthesis of ketoprofen is preferable because of its mild reaction conditions and eco-friendliness.In this review,the progress in biosynthesis of chiral ketoprofen with lipase,esterase,and nitrile hydratase/amidase bienzyme system as biocatalysts was summarized.
