摘要

胞苷二磷酸胆碱(CDPC)是一种有着广阔市场的原料药,目前国内最先进的生产CDPC的方法是酵母发酵法。这种方法具有菌种用量少、转化率高、产品质量好的优点。灌流培养是一种利用物料循环和切向过滤的原理,将生物发酵产物源源不断分离出来的方法。为了弥补普通发酵法产能有限和设备利用率较低的不足,将灌流法应用到CDPC 的生产工艺中。结果显示灌流法生产CDPC 的产能是非灌流法的3.96 倍,同时CDPC 溶液色度(A430值)与非灌流法相比,降低了77.7%,产品质量显著提高。灌流法生产CDPC 能够有效提高产品的质量和产能,提高设备的利用率,同时大幅降低了生产成本。

Abstract

Cytidine Diphosphate Choline (CDPC) is an effective pharmaceutical ingredient with broad market prospect. Currently, yeast fermentation is the most advanced production method for CDPC in China, which has advantages of less consumption of culture, high conversion rate and good product quality. The process of perfusion culture can separate the fermentation products continuously based on material circulation and tangential filtration. In order to solve the disadvantage
of limited production capacity and low efficiency of equipment utilization, a new perfusion system was applied in CDPC biosynthesis. The results showed that the production capacity was 3.96 times higher compare with that of non-perfusion method and the color degree of CDPC solution (A430) reduced
by 77.7%. The application of perfusion method in CDPC production effectively improved the product quality, production capacity and equipment utilization, and meanwhile significantly reduced
the production cost.
