摘要

研究四氮唑盐/甲臜类EZMTT法检测细胞活力和增殖的应用并评价EZMTT法的优势.评价EZMTT法检测细胞增殖的准确性,对比EZMTT法与WST-8的灵敏度、毒性及细胞活力长期跟踪的应用.研究表明:细胞量为3 000~500 000时,在4h内EZMTT法信号值与细胞量成正比,且较现有产品WST-8检测试剂,EZMTT法更灵敏、毒性更低,可实现长达7d的细胞活力跟踪.0.06%十二烷基硫酸钠或50μmol/L盐酸可中止EZMTT反应.EZMTT法有望成为更为优秀的新一代细胞活力检测方法.

Abstract

This study focused on the application of EZMTT assay based on tetrazolium salts/formazans reaction for cell viability,as well as its superiority evaluation.The accuracy of  EZMTT assay was evaluated on efficiency and toxicity.When the cell number is 3 000to 500 000, the linear dose response can be achieved within 4hours.The EZMTT assay exhibited higher  sensibility and effectiveness compared with WST-8assay in cell viability test.Besides,the low  toxicity allows EZMTT track cell growth in long-term,and the EZMTT reaction could be   stopped by 0.06%SDS or 50μmol/L HCL.The high efficiency and low toxicity demonstrated  the great potential of EZMTT assay for cell viability test.
